    Phonological issues in the production of prosody by francophone and sinophone learners of english as a second language

    Un accent de non-natif peut mener à une incompréhension ou à la perception de degrés différents d'accent d'étrangeté. La prosodie, qui est maintenant reconnue comme un élément important de l'impression d'étrangeté, est relativement peu abordée en recherche en acquisition des langues étrangères. Ceci contraste avec l'intérêt grandissant envers la prosodie en tant qu'élément de la langue maternelle. Dans cette thèse, la recherche phonologique est évaluée quant à sa pertinence dans la recherche sur la prosodie des langues étrangères. Deux aspects de la théorie phonologique sont étudiés: la typologie et l'organisation phonologique. Ce choix est justifié par la présomption générale que l'étrangeté prosodique est créée soit par une différence de typologie entre langue maternelle (L1) et langue étrangère (L2) soit par un transfert de traits prosodiques de la L1. La critique de la recherche en typologie phonologique conclut que, à ce stade, aucun modèle de classification prosodique n'est applicable à l'acquisition d'une L2. En particulier, l'étude démontre que certaines typologies, en particulier la théorie de l'isochronie accentuelle/l'isochronie syllabique de Pike, devraient être exclues parce qu'elles entravent les progrès en recherche sur l'acquisition et la production de la prosodie des langues étrangères. Le second aspect de la théorie phonologique étudié dans cette thèse est l'organisation phonologique. La prémisse est que les différences sous-jacentes à l'organisation prosodique plutôt que les différences phonologiques de surface sont transférées de L1 à L2. Les analyses approfondies de l'anglais nord américain, le français et le chinois standard révèlent d'importantes différences phonologiques entre l'anglais nord américain et les deux autres langues. Quatre expériences évaluent certaines de ces différences. La prosodie de l'anglais produite par des locuteurs natifs du français est analysée dans des phrases rythmiquement simples et des phrases rythmiquement plus complexes. Les résultats démontrent que l'accentuation lexicale est moins problématique que l'accentuation prosodique supra-lexicale. En particulier, il est démontré que les montées de fréquence fondamentale (F0) de début et de fin de syntagme accentuel (SA), typiques du français, sont source d'erreur dans la prosodie de l'anglais langue seconde. Il est cependant montré que cette erreur, bien que remarquée par les locuteurs natifs de l'anglais, n'affecte pas la perception de placement d'accentuation par ces derniers. La prosodie de l'anglais produite par des locuteurs natifs du chinois est analysée en termes de transfert de ton et d'alignement de pic de F0. Les résultats indiquent que les locuteurs du chinois utilisent les tons chinois quand ils produisent des tons accentuels de l'anglais; plus spécifiquement, la majorité des locuteurs utilisent le ton 2 (ton montant) quand ils produisent un ton accentuel montant. La dernière expérience révèle que les locuteurs natifs du chinois alignent le ton accentuel avec la syllabe accentuée à laquelle elle correspond de manière plus stricte que les locuteurs natifs de l'anglais nord américain le font. Les résultats de cette thèse génèrent un aperçu de la progression de la performance de la prosodie d'une langue étrangère.     Developing the first pan-Canadian survey on patient engagement in patient safety

    Abstract Background Patient safety is a worldwide problem, and the patient contribution to mitigate the risk of patient harm is now recognized as a cornerstone to its solution. In order to understand the nature of integrating patients into patient safety and healthcare organizations and to monitor their integration, a Canadian survey tool has been co-constructed by patients, researchers and the Canadian Patient Safety Institute (CPSI). This questionnaire has been adapted from the French version of the patient engagement (PE) in patient safety (PS) questionnaire created for the province of Quebec, Canada. Methodology The pan-Canadian PE in PS survey tool was developed in a five-step process: (1) a literature review and revision of the initial tool developed in the province of Quebec; (2) translation of the French questionnaire into an English version tool; (3) creation of a Canadian expert advisory group; (4) adaptation of the English version tool based on feedback from the expert advisory group (assessment and development of the construct’s dimensions, wording assessment and adaptation for pan-Canadian use, technical testing of the online platform for the survey); and (5) pilot testing and pre-validation of the tool before pan-Canadian use. Results and conclusion Eight pan-Canadian PE in PS surveys were completed from five Canadian provinces by the expert advisory group and six surveys were completed during the pilot project by participants from different provinces in Canada. This survey tool comprises 5 sections: (1) demographic identification of the participants (Q1 to Q5); (2) general questions (Q6 to Q17); (3) the patient engagement process (experience level of participants and organizational incentives for PE in general) (Q18 to Q33); (4) PE in PS processes, such as current activities, strategies, structures, resources and factors (Q34 to Q67); and (5) the context and impact of PE in PS initiatives in Canadian healthcare organizations (CHOs) (Q68 to Q75), including outcome identification, improvement mechanisms and strategies, evaluation mechanisms, and indicators

    Detection of novel recombinases in bacteriophage genomes unveils Rad52, Rad51 and Gp2.5 remote homologs

    Homologous recombination is a key in contributing to bacteriophages genome repair, circularization and replication. No less than six kinds of recombinase genes have been reported so far in bacteriophage genomes, two (UvsX and Gp2.5) from virulent, and four (Sak, Redβ, Erf and Sak4) from temperate phages. Using profile–profile comparisons, structure-based modelling and gene-context analyses, we provide new views on the global landscape of recombinases in 465 bacteriophages. We show that Sak, Redβ and Erf belong to a common large superfamily adopting a shortcut Rad52-like fold. Remote homologs of Sak4 are predicted to adopt a shortcut Rad51/RecA fold and are discovered widespread among phage genomes. Unexpectedly, within temperate phages, gene-context analyses also pinpointed the presence of distant Gp2.5 homologs, believed to be restricted to virulent phages. All in all, three major superfamilies of phage recombinases emerged either related to Rad52-like, Rad51-like or Gp2.5-like proteins. For two newly detected recombinases belonging to the Sak4 and Gp2.5 families, we provide experimental evidence of their recombination activity in vivo. Temperate versus virulent lifestyle together with the importance of genome mosaicism is discussed in the light of these novel recombinases.     Use of ChAd3-EBO-Z Ebola virus vaccine in Malian and US adults, and boosting of Malian adults with MVA-BN-Filo: a phase 1, single-blind, randomised trial, a phase 1b, open-label and double-blind, dose-escalation trial, and a nested, randomised, double-blind, placebo-controlled trial

    SummaryBackgroundThe 2014 west African Zaire Ebola virus epidemic prompted worldwide partners to accelerate clinical development of replication-defective chimpanzee adenovirus 3 vector vaccine expressing Zaire Ebola virus glycoprotein (ChAd3-EBO-Z). We aimed to investigate the safety, tolerability, and immunogenicity of ChAd3-EBO-Z in Malian and US adults, and assess the effect of boosting of Malians with modified vaccinia Ankara expressing Zaire Ebola virus glycoprotein and other filovirus antigens (MVA-BN-Filo).MethodsIn the phase 1, single-blind, randomised trial of ChAd3-EBO-Z in the USA, we recruited adults aged 18–65 years from the University of Maryland medical community and the Baltimore community. In the phase 1b, open-label and double-blind, dose-escalation trial of ChAd3-EBO-Z in Mali, we recruited adults 18–50 years of age from six hospitals and health centres in Bamako (Mali), some of whom were also eligible for a nested, randomised, double-blind, placebo-controlled trial of MVA-BN-Filo. For randomised segments of the Malian trial and for the US trial, we randomly allocated participants (1:1; block size of six [Malian] or four [US]; ARB produced computer-generated randomisation lists; clinical staff did randomisation) to different single doses of intramuscular immunisation with ChAd3-EBO-Z: Malians received 1 × 1010 viral particle units (pu), 2·5 × 1010 pu, 5 × 1010 pu, or 1 × 1011 pu; US participants received 1 × 1010 pu or 1 × 1011 pu. We randomly allocated Malians in the nested trial (1:1) to receive a single dose of 2 × 108 plaque-forming units of MVA-BN-Filo or saline placebo. In the double-blind segments of the Malian trial, investigators, clinical staff, participants, and immunology laboratory staff were masked, but the study pharmacist (MK), vaccine administrator, and study statistician (ARB) were unmasked. In the US trial, investigators were not masked, but participants were. Analyses were per protocol. The primary outcome was safety, measured with occurrence of adverse events for 7 days after vaccination. Both trials are registered with ClinicalTrials.gov, numbers NCT02231866 (US) and NCT02267109 (Malian).FindingsBetween Oct 8, 2014, and Feb 16, 2015, we randomly allocated 91 participants in Mali (ten [11%] to 1 × 1010 pu, 35 [38%] to 2·5 × 1010 pu, 35 [38%] to 5 × 1010 pu, and 11 [12%] to 1 × 1011 pu) and 20 in the USA (ten [50%] to 1 × 1010 pu and ten [50%] to 1 × 1011 pu), and boosted 52 Malians with MVA-BN-Filo (27 [52%]) or saline (25 [48%]). We identified no safety concerns with either vaccine: seven (8%) of 91 participants in Mali (five [5%] received 5 × 1010 and two [2%] received 1 × 1011 pu) and four (20%) of 20 in the USA (all received 1 × 1011 pu) given ChAd3-EBO-Z had fever lasting for less than 24 h, and 15 (56%) of 27 Malians boosted with MVA-BN-Filo had injection-site pain or tenderness.Interpretation1 × 1011 pu single-dose ChAd3-EBO-Z could suffice for phase 3 efficacy trials of ring-vaccination containment needing short-term, high-level protection to interrupt transmission.     Cooperation of breast cancer proteins PALB2 and piccolo BRCA2 in stimulating homologous recombination.

    Inherited mutations in human PALB2 are associated with a predisposition to breast and pancreatic cancers. PALB2's tumor-suppressing effect is thought to be based on its ability to facilitate BRCA2's function in homologous recombination. However, the biochemical properties of PALB2 are unknown. Here we show that human PALB2 binds DNA, preferentially D-loop structures, and directly interacts with the RAD51 recombinase to stimulate strand invasion, a vital step of homologous recombination. This stimulation occurs through reinforcing biochemical mechanisms, as PALB2 alleviates inhibition by RPA and stabilizes the RAD51 filament. Moreover, PALB2 can function synergistically with a BRCA2 chimera (termed piccolo, or piBRCA2) to further promote strand invasion. Finally, we show that PALB2-deficient cells are sensitive to PARP inhibitors. Our studies provide the first biochemical insights into PALB2's function with piBRCA2 as a mediator of homologous recombination in DNA double-strand break repair

    Prosodic Transfer: From Chinese Lexical tone to English Pitch Accent

    Chinese tones are associated with a syllable to convey meaning, English pitch accents are prominence markers associated with stressed syllables.  As both are created by pitch modulation, their pitch contours can be quite similar.  The experiment reported here examines whether native speakers of Chinese produce, when speaking English, the Chinese tone whose phonetic contour most closely matches the contour of the intended English pitch accent.  Six native speakers of Chinese recorded English and Chinese sentences, all including the segment [fan].  Results show that the subjects produced a Chinese tone 2 where a rising pitch accents was required and thus that speakers of Chinese rely on their lexical tones inventory to produce English prosody. The results obtained with falling pitch accents are much less conclusive partly because of the difficulty in measuring tone 3 due to the high level of creak that accompanies it
